也將細(xì)胞數(shù)量擴(kuò)大，就必須進(jìn)行傳代(再培養(yǎng))。 傳代培養(yǎng)也是一種將細(xì)胞種保存下去的方法。同時(shí)也是利用培養(yǎng)細(xì)胞進(jìn)行各種實(shí)驗(yàn)的必經(jīng)過(guò)程。懸浮型細(xì)胞直接分瓶就可以，而貼壁細(xì)胞需經(jīng)消化后才能分瓶。

1、冷凍管應(yīng)如何解凍？
取出冷凍管后， 須立即放入37 ℃水槽中快速解凍， 輕搖冷凍管使其在1 分鐘內(nèi)全部融化， 并注意水面不可超過(guò)冷凍管蓋沿， 否則易發(fā)生污染情形。另冷凍管由液氮桶中取出解凍時(shí)， 必須注意安全， 預(yù)防人卵巢畸胎瘤細(xì)胞；PA-1培養(yǎng)冷凍管之爆裂。
2、細(xì)胞冷凍管解凍培養(yǎng)時(shí)， 是否應(yīng)馬上去除DMSO？
除少數(shù)特別注明對(duì)DMSO 敏感之細(xì)胞外， 絕大部分細(xì)胞株（包括懸浮性細(xì)胞）， 在解凍之后， 應(yīng)直接放入含有10-15ml新鮮培養(yǎng)基之培養(yǎng)角瓶中， 待隔天再置換新鮮培養(yǎng)基以去除DMSO 即可， 如此可避免大部分解凍后細(xì)胞無(wú)法生長(zhǎng)或貼附之問(wèn)題。
3、可否使用與原先培養(yǎng)條件不同之培養(yǎng)基？
不能。每一細(xì)胞株均有其特定使用且已適應(yīng)之細(xì)胞培養(yǎng)基， 若驟然使用和原先提供之培養(yǎng)條件不同之培養(yǎng)基， 細(xì)胞大都無(wú)法立即適應(yīng)， 造成細(xì)胞無(wú)法存活。
4、可否使用與原先培養(yǎng)條件不同之血清種類？
不能。血清是細(xì)胞培養(yǎng)上一個(gè)極為重要的營(yíng)養(yǎng)來(lái)源， 所以血清的種類和品質(zhì)對(duì)于細(xì)胞的生長(zhǎng)會(huì)產(chǎn)生*的影響。來(lái)自不同物種的血清， 在一些物質(zhì)或分子的量或內(nèi)容物上都有所不同，血清使用錯(cuò)誤常會(huì)造成細(xì)胞無(wú)法存活。
人卵巢畸胎瘤細(xì)胞；PA-1培養(yǎng)注意事項(xiàng)：
1. 收到細(xì)胞后首先觀察細(xì)胞瓶是否完好，培養(yǎng)液是否有漏液、渾濁等現(xiàn)象，若有上述現(xiàn)象發(fā)生請(qǐng)及時(shí)和我們。
2. 仔細(xì)閱讀細(xì)胞說(shuō)明書(shū)，了解細(xì)胞相關(guān)信息，如細(xì)胞形態(tài)、所用培養(yǎng)基、血清比例、所需細(xì)胞因子等。
3. 用75%酒精擦拭細(xì)胞瓶表面，顯微鏡下觀察細(xì)胞狀態(tài)。因運(yùn)輸問(wèn)題貼壁細(xì)胞會(huì)有少量從瓶壁脫落，將細(xì)胞置于培養(yǎng)箱內(nèi)靜置培養(yǎng)，隔天再取出觀察。此時(shí)多數(shù)細(xì)胞均會(huì)貼壁，若細(xì)胞仍不能貼壁請(qǐng)用臺(tái)盼藍(lán)染色測(cè)定細(xì)胞活力，如果證實(shí)細(xì)胞活力正常，請(qǐng)將細(xì)胞離心后用新鮮培養(yǎng)基再次貼壁培養(yǎng)；如果染色結(jié)果顯示細(xì)胞無(wú)活力，請(qǐng)拍下照片及時(shí)和我們，信息確認(rèn)后我們?yōu)槟倜赓M(fèi)寄送一次。
4. 請(qǐng)客戶用相同條件的培養(yǎng)基用于細(xì)胞培養(yǎng)。培養(yǎng)瓶?jī)?nèi)多余的培養(yǎng)基可收集備用，細(xì)胞傳代時(shí)可以一定比例和客戶自備的培養(yǎng)基混合，使細(xì)胞逐漸適應(yīng)培養(yǎng)條件。
5. 建議客戶收到細(xì)胞后前3天各拍幾張細(xì)胞照片，記錄細(xì)胞狀態(tài)，便于和公司技術(shù)部溝通交流。
6. 該細(xì)胞只能用于科研，不得用于臨床應(yīng)用?！加猛尽剑荷芯?。沉淀鉀、銣、銫、銨，以與鈉分離。

產(chǎn)品介紹 GNL2 is a nucleolar guanasine-triphosphate binding protein that is ubiquitously expressed at low levels in almost all tissues. GNL2 is involved in the crucial process of trafficking proteins out of the nucleus. Specifically, it is a GTPase that interacts with the 60s preribosomal subunit in the nucleus and facilitates export of the subunit into the cytoplasm. GTPases are responsible for the hydrolysis of GTP by way of a protein region dubbed the G domain. GTPases are often involved in the translocating proteins through membranes gleaning energy for the activity by hydrolizing GTP. GNL2 shares G domain homology and some functionality with nucleostemin (GNL3), another nuclear GTPase. Highest expression of GNL2 is found in testis.
Function : GTPase that associates with pre-60S ribosomal subunits in the nucleolus and is required for their nuclear export and maturation.
Subcellular Location : Nucleus; nucleolus.

產(chǎn)品介紹 GTPases from the MMR1/HSR1 GTP-binding protein subfamily are circularly rearranged G-motifs that play a critical role in maintaining normal cell growth. Deletion of these genes results in severe growth defects with a marked reduction in mature rRNA species and a concomitant accumulation of the 35S pre-rRNA transcript. Deletion also causes the ribosomal protein Rpl25a to fail exportation from the nucleolus. Deletion of any of the G-domain motifs will result in a null phenotype and nuclear/nucleolar localization that lacks the nucleolar export of preribosomes and is accompanied by a distortion of the nucleolar structure. GNL3L (guanine nucleotide binding protein-like 3 (nucleolar)-like) is a 582 amino acid nuclear protein that belongs to the MMR1/HSR1 GTP-binding protein family. Containing one G (guanine nucleotide-binding) domain, GNL3L is required for normal processing of ribosomal pre-rRNA and for cell proliferation.
Function : Stabilizes TERF1 omeric association by preventing TERF1 recruitment by PML. Stabilizes TERF1 protein by preventing its ubiquitination and hence proteasomal degradation. Does so by interfering with TERF1-binding to FBXO4 E3 ubiquitin-protein ligase. Required for cell proliferation. By stabilizing TRF1 protein during mitosis, promotes metaphase-to-anaphase transition.
