1.實驗之前準備好所有的標準品（見備用試劑）。建議所有的標準和樣本都做復孔。

2.首先把微孔板固定在支架上，分別設好相應的孔（空白孔（空白對照孔不加樣品及酶標試劑，其余各步操作相同）、標準孔、待測樣品孔），然后在包被板上，標準品和代測樣本孔準確加樣50μl

3.加10 μL的*到代測樣本孔中。（標準孔不加）
4.每孔加入酶標試劑50μl，混合，混合在這一步是很重要的。封板后置37℃溫育1個小時。
5.在溫育結束前15分鐘內(nèi)準備顯色液。（見備用試劑）
6.用下面的一種方法洗板;
7.手洗：將微孔板中的混合物倒入合適的廢物器皿中，然后每孔注滿稀釋好的洗滌液，再到掉，如此重復4次，然后倒置微孔板，將其拍到吸水紙上或者紙巾上，直到拍干。、
（注：洗板的時候要緊握微孔板的兩側，以確保所有的板條不脫落）
8.機洗：吸干所有的孔，然后用洗板機洗板5次，調(diào)整洗板機盡可能洗掉所有的液體，設置填充量在350微升/孔/清洗（范圍：350-400微升），洗板后，倒置微孔板，將其拍到吸水紙上或者紙巾上，直到拍干。建議洗板機設置的浸泡時間為10秒，震動時間為5秒。
9.每孔依次加入顯色劑A 50μl，顯色劑B 50μl，封板后置20-25°C顯色15分鐘.
10.每孔加終止液50μl混合
11.在30分鐘以內(nèi)讀酶標儀在450nm波長的O.D值
