描述 | OP9細(xì)胞株源自新生的op/op 小鼠顱蓋。因編碼M-CSF的基因中的一個突變,它不能生成有功能的巨噬細(xì)胞克隆刺激因子(M-CSF)。M-CSF的存在對胚胎干細(xì)胞(ES)分化成血細(xì)胞而不是其他巨噬細(xì)胞有抑制功能。 OP9細(xì)胞可以用于與小鼠胚胎干細(xì)胞共培養(yǎng)以誘導(dǎo)胚胎干細(xì)胞分化成成紅血球來源的、骨髓來源的和B細(xì)胞譜系的血細(xì)胞。與OP9共培養(yǎng)不需要外源的生長因子或復(fù)雜的胚胎結(jié)構(gòu)。這個系統(tǒng)對研究造血細(xì)胞的發(fā)育和分化的分子機理有用。 |
