用法:血清由冰凍狀態(tài)在常溫下*融化成液體,搖均勻后放置10-30分鐘待結(jié)塊蛋白質(zhì)沉底后即可使用。
注意事項(xiàng):有少量蛋白質(zhì)結(jié)塊析出不影響使用。未融化或融化未經(jīng)搖勻的血清禁止直接放入高溫水浴內(nèi)加溫。減少血清重復(fù)凍融。血清在室溫或4℃冰箱內(nèi)放置過久會(huì)影響促細(xì)胞生*果。
的保存方法:
(1)*保存的血清必須儲(chǔ)存于-20℃ - 70℃ 低溫冰箱中.4℃冰箱中保存時(shí)間切勿超過1個(gè)月.由于血清結(jié)冰時(shí)體積會(huì)增加約10%,因此,血清在凍入低溫冰箱前,必須預(yù)留一定體積空間,否則易發(fā)生污染或玻璃瓶凍裂.
(2)一般廠商提供的血清為無菌,無需再過濾除菌.如發(fā)現(xiàn)血清有懸浮物,則可將血清加入培養(yǎng)液內(nèi)一起過濾,切勿直接過濾血清.
(3)血清解凍: 瓶裝血清解凍需采用逐步解凍法:-20℃ 至 -70℃ 低溫冰箱中的血清放入4℃冰箱中溶解1天.然后移入室溫,待全部溶解后再分裝.在溶解過程中需不斷輕輕搖晃均勻(小心勿造成氣泡),使溫度與成分均一,減少沉淀的發(fā)生.切勿直接將血清從-20℃進(jìn)入37℃解凍,這樣因溫度改變太大,容易造成蛋白質(zhì)凝集而出現(xiàn)沉淀。
