背景介紹：GALR3 a 368 and 370 amino acid protein in human and rat, respectively, belongs to a family of G protein-coupled receptors that bind the neuropeptide galanin, which is distributed throughout the central and peripheral nervous system, the pituitary gland, the gastrointestinal tract and in the endocrine and exocrine pancreas. GALR3 mRNA is widely distributed, but expressed at low abundance. In human, GALR3 mRNA is highly expressed in the hypothalamus, pituitary and testis, and is expressed to a lesser extent in adrenal gland and pancreas. Rat and human GALR3 co-express with potassium channel subunits GIRK1 and GIRK4. Like GALR1, GALR3 signaling pathways lead to the inhibition of adenylate cyclase and to the activation of potassium channels, which are linked to the regulation of neurotransmitter release. Binding of galanin to galanin receptors results in increased feeding, impaired learning, enhanced opiate analgesia and decreased opiate place preference.

甘丙肽受體3抗體保存條件: GLRB (Glycine receptor beta) is a neurotransmitter-gated ion channel concentrated within the spinal cord and brainstem. Expression is also observed in several upper brain regions including the cortex, cerebellum, hippocampus and amygdala. Binding of glycine to GLRB increases the chloride conductance and thus produces hyperpolarization (inhibition of neuronal firing), controlling spinal reflexes and locomotor behavior.

石蠟切片的免疫熒光單染(間接法)

1.石蠟切片正常脫蠟至水，PBS洗5分鐘，2次
2.抗原修復，0.01M枸櫞酸修復液，沸水浴熱修復15分鐘
3.正常山羊血清封閉20分鐘，37℃
4.一抗孵育，4℃過ye。
5.PBS洗5分鐘，3次
6.熒光標記二抗孵育，37℃，90分鐘
7.PBS洗5分鐘，3次
8.DAPI復染，PBS洗5分鐘，3次
9.防淬滅熒光封片劑封片

甘丙肽受體3抗體,Anti-GALR3抗體石蠟切片的免疫組化SP法參考步驟：

1.60℃烤片2小時
2.脫蠟。二甲苯脫蠟15分鐘，三次
3.水化。依次經過無水乙醇、95%、90%、80%、70%，各5分鐘，蒸餾水（或自來水）5分鐘
4.PBS洗5分鐘，三次
5.抗原修復。枸櫞酸緩沖液（約92-95℃）煮沸（沸水浴）修復15分鐘，自然涼至室溫。PBS洗5分鐘，三次
6.3%雙氧水消化內源性的過氧化物酶，20分鐘，PBS洗5分鐘，三次
7.正常山羊血清封閉，37℃20分鐘
8.一抗孵育，4℃過ye.
9.PBS洗5分鐘，三次
10.二抗孵育，37℃20分鐘（二抗為*標記的羊抗兔IgG），PBS洗5分鐘，三次
11.三抗孵育，37℃20分鐘（三抗為辣根酶標記的鏈霉親和素），PBS洗5分鐘，三次
12.DAB顯色，鏡下觀察結果，適時終止
13.蘇木素復染5分鐘，自來水沖洗片刻，1%的鹽酸酒精（75%）溶液（請自行配置，配制比例為：100ml 75%酒精+1ml濃鹽酸）分化30秒，自來水沖洗5分鐘藍化
14.脫水。依次經過70%、80%、90%、95%、無水乙醇，各5分鐘，二甲苯15分鐘，三次
15.封片，中性樹膠封片
