一 開機(jī)前檢查：
1 檢查儀器臺(tái)面（DECK）上所有的實(shí)驗(yàn)材料（Labware）。
2 檢查SystemWater 水桶的水位。
3 檢查恒溫循環(huán)水浴（Chiller）水箱的水位，并定期更換或填充Chiller 中的循環(huán)水。
4 倒掉廢液桶中的廢液。
二 開機(jī)步驟：
打開Chiller、Heater、儀器及計(jì)算機(jī)電源，進(jìn)入WinPREP程序 ：
1 儀器初始化
點(diǎn)擊菜單：[Utilites]→[Setup]→[Instrument]→[Initialize]儀器進(jìn)行初始化。
2 系統(tǒng)管路沖洗、除氣泡
點(diǎn)擊菜單：[Utilites]→[Diagnostics Test]→[Flush and Wash tips]；
放置Labware(washbowl+1Through)在儀器平臺(tái)的G13位置，排液管接至廢液桶，按提示執(zhí)行程序：
Deck layout OK,
run test now。 √
Flush Wash Volumes
Enter the Flush Volume(vl): 3000
Enter the Wash Volume(vl) : 3000
>start,儀器開始沖洗管路中的氣泡，要多次沖洗直至管中*沒有氣泡。
3 執(zhí)行自動(dòng)酶切程序
點(diǎn)擊菜單：[File]→[Open]；
打開所要執(zhí)行的自動(dòng)酶切操作文件：
按文件圖示位置放置所需的Labware。（注意：蓋子應(yīng)放在96 well ABGENE PCR 上； 所有實(shí)驗(yàn)器皿應(yīng)放入準(zhǔn)確位置）， 按要求配置試劑，裝入試劑盒（瓶）；
點(diǎn)擊工具欄：[Execute Test]。彈出[Test Startup page]窗口, →點(diǎn)擊[Reset Tip Boxes],（Lid on PCR(1-96) ,Tips=1），“Fill”為添滿，“Empty”為清空，

>Start，開始執(zhí)行程序。
注意事項(xiàng)：
1． 為避免角蛋白和其它污染，在移動(dòng)labware 、dispensable tips、試劑或樣品時(shí)，請(qǐng)戴手套操作，在儀器運(yùn)行過程中非儀器操作人員請(qǐng)遠(yuǎn)離儀器。
2． 整個(gè)操作過程中的水為除去氣泡的去離子水。
3． 檢查冷卻循環(huán)儀是否打開和是否達(dá)到安全水位；檢查系統(tǒng)液體的水平面是否達(dá)到標(biāo)志線（1/2桶的體積）；檢查廢水桶的液面，及時(shí)排水；檢查各種實(shí)際是否準(zhǔn)確配置及正確放入位置。
4． 執(zhí)行程序前，一定要對(duì)照所選執(zhí)行程序檢查所有Labware的位置是否正確
5． 蓋子（lid）應(yīng)放在96 well ABgene PCR （1-96）上。
6． 運(yùn)行FlushLiq ，盡可能的去除系統(tǒng)中可能存在的氣泡。
7． 在[Reset Tip Boxes]時(shí)，要確定蓋子（lid）位置和槍頭（Tips）數(shù)量與實(shí)際位置和數(shù)量相符
8． 在儀器運(yùn)行過程中如有以外情況發(fā)生，請(qǐng)速按儀器正前方緊急停止按扭（紅色）。
9． 在儀器關(guān)機(jī)不用時(shí)，請(qǐng)用儀器罩遮嚴(yán)以防止灰塵。
10． 在每次操作完成后請(qǐng)自覺登記，并保證操作臺(tái)面的整潔。
