也將細(xì)胞數(shù)量擴(kuò)大，就必須進(jìn)行傳代(再培養(yǎng))。 傳代培養(yǎng)也是一種將細(xì)胞種保存下去的方法。同時(shí)也是利用培養(yǎng)細(xì)胞進(jìn)行各種實(shí)驗(yàn)的必經(jīng)過(guò)程。懸浮型細(xì)胞直接分瓶就可以，而貼壁細(xì)胞需經(jīng)消化后才能分瓶。

1、冷凍管應(yīng)如何解凍？
取出冷凍管后， 須立即放入37 ℃水槽中快速解凍， 輕搖冷凍管使其在1 分鐘內(nèi)全部融化， 并注意水面不可超過(guò)冷凍管蓋沿， 否則易發(fā)生污染情形。另冷凍管由液氮桶中取出解凍時(shí)， 必須注意安全， 預(yù)防冷凍管之爆裂。
2、細(xì)胞冷凍管解凍培養(yǎng)時(shí)， 是否應(yīng)馬上去除DMSO？
除少數(shù)特別注明對(duì)DMSO 敏感之細(xì)胞外， 絕大部分細(xì)胞株（包括懸浮性細(xì)胞）， 在解凍之后， 應(yīng)直接放入含有10-15ml新鮮培養(yǎng)基之培養(yǎng)角瓶中， 待隔天再置換新鮮培養(yǎng)基以去除DMSO 即可， 如此可避免大部分解凍后細(xì)胞無(wú)法生長(zhǎng)或貼附之問(wèn)題。
3、可否使用與原先培養(yǎng)條件不同之培養(yǎng)基？
人前列腺癌細(xì)胞；DU 145 [DU145；DU-145]培養(yǎng)不能。每一細(xì)胞株均有其特定使用且已適應(yīng)之細(xì)胞培養(yǎng)基， 若驟然使用和原先提供之培養(yǎng)條件不同之培養(yǎng)基， 細(xì)胞大都無(wú)法立即適應(yīng)， 造成細(xì)胞無(wú)法存活。
4、可否使用與原先培養(yǎng)條件不同之血清種類(lèi)？
不能。血清是細(xì)胞培養(yǎng)上一個(gè)極為重要的營(yíng)養(yǎng)來(lái)源， 所以血清的種類(lèi)和品質(zhì)對(duì)于細(xì)胞的生長(zhǎng)會(huì)產(chǎn)生*的影響。來(lái)自不同物種的血清， 在一些物質(zhì)或分子的量或內(nèi)容物上都有所不同，血清使用錯(cuò)誤常會(huì)造成細(xì)胞無(wú)法存活。
注意事項(xiàng)：
1. 收到細(xì)胞后首先觀察細(xì)胞瓶是否完好，培養(yǎng)液是否有漏液、渾濁等現(xiàn)象，若有上述現(xiàn)象發(fā)生請(qǐng)及時(shí)和我們。
2. 仔細(xì)閱讀細(xì)胞說(shuō)明書(shū)，了解細(xì)胞相關(guān)信息，如細(xì)胞形態(tài)、所用培養(yǎng)基、血清比例、所需細(xì)胞因子等。
3. 用75%酒精擦拭細(xì)胞瓶表面，顯微鏡下觀察細(xì)胞狀態(tài)。因運(yùn)輸問(wèn)題貼壁細(xì)胞會(huì)有少量從瓶壁脫落，將細(xì)胞置于培養(yǎng)箱內(nèi)靜置培養(yǎng)，隔天再取出觀察。此時(shí)多數(shù)細(xì)胞均會(huì)貼壁，若細(xì)胞仍不能貼壁請(qǐng)用臺(tái)盼藍(lán)染色測(cè)定細(xì)胞活力，如果證實(shí)細(xì)胞活力正常，請(qǐng)將細(xì)胞離心后用新鮮培養(yǎng)基再次貼壁培養(yǎng)；如果染色結(jié)果顯示細(xì)胞無(wú)活力，請(qǐng)拍下照片及時(shí)和我們，信息確認(rèn)后我們?yōu)槟倜赓M(fèi)寄送一次。
4. 請(qǐng)客戶(hù)用相同條件的培養(yǎng)基用于細(xì)胞培養(yǎng)。培養(yǎng)瓶?jī)?nèi)多余的培養(yǎng)基可收集備用，細(xì)胞傳代時(shí)可以一定比例和客戶(hù)自備的培養(yǎng)基混合，使細(xì)胞逐漸適應(yīng)培養(yǎng)條件。
5. 建議客戶(hù)收到細(xì)胞后前3天各拍幾張細(xì)胞照片，記錄細(xì)胞狀態(tài)，便于和公司技術(shù)部溝通交流。
6. 該細(xì)胞只能用于科研，不得用于臨床應(yīng)用?！加猛尽剑荷芯俊３恋礅?、銣、銫、銨，以與鈉分離。

產(chǎn)品介紹 This gene encodes two of three subunit types of the membrane-bound enzyme N-acetylglucosamine-1-phosphotransferase, a heterohexameric complex composed of two alpha, two beta, and two gamma subunits. The encoded protein is proteolytically cleaved at the Lys928-Asp929 bond to yield mature alpha and beta polypeptides while the gamma subunits are the product of a distinct gene (GeneID 84572). In the Golgi apparatus, the heterohexameric complex catalyzes the first step in the synthesis of mannose 6-phosphate recognition markers on certain oligosaccharides of newly synthesized lysosomal enzymes. These recognition markers are essential for appropriate trafficking of lysosomal enzymes. Mutations in this gene have been associated with both mucolipidosis II and mucolipidosis IIIA.[provided by RefSeq, May 2010].
Function : Catalyzes the formation of mannose 6-phosphate (M6P) markers on high mannose type oligosaccharides in the Golgi apparatus. M6P residues are required to bind to the M6P receptors (MPR), which mediate the vesicular transport of lysosomal enzymes to the endosomal/prelysosomal compartment.
Subunit : Hexamer of two alpha, two beta and two gamma subunits; disulfide-linked. It is believed that the alpha and/or the beta subunit of the enzyme contain the catalytic portion and that the gamma subunit functions in recognition of the lysosomal enzymes.

DISEASE : Defects in GNPTAB are the cause of mucolipidosis type II (MLII) [MIM:252500]; also known as inclusion cell disease or I-cell disease (ICD). MLII is a fatal, autosomal recessive, lysosomal storage disorder characterized by severe clinical and radiologic features, peculiar fibroblast inclusions, and no excessive mucopolysacchariduria. Congenital dislocation of the hip, thoracic deformities, hernia, and hyperplastic gums are evident soon after birth.
Defects in GNPTAB are the cause of mucolipidosis type III complementation group A (MLIIIA) [MIM:252600]; also known as variant pseudo-Hurler polydystrophy. MLIIIA is an autosomal recessive disease of lysosomal enzyme targeting. Clinically MLIII is characterized by restricted joint mobility, skeletal dysplasia, and short stature. Mildly coarsened facial features and thickening of the skin have been described. Cardiac valvular disease and corneal clouding may also occur. Half of the reported patients show learning disabilities or mental retardation.

DISEASE : Defects in GNPTG are the cause of mucolipidosis type III complementation group C (MLIIIC) [MIM:252605]; also known as variant pseudo-Hurler polydystrophy. MLIIIC is an autosomal recessive disease of lysosomal hydrolase trafficking. Unlike the related diseases, mucolipidosis II and IIIA, the enzyme affected in mucolipidosis IIIC (GlcNAc-phosphotransferase) retains full transferase activity on synthetic substrates but lacks activity on lysosomal hydrolases. Typical clinical findings include stiffness of the hands and shoulders, claw-hand deformity, scoliosis, short stature, coarse facies, and mild mental retardation. Radiographically, severe dysostosis multiplex of the hip is characteristic and frequently disabling. The clinical diagnosis can be confirmed by finding elevated serum lysosomal enzyme levels and/or decreased lysosomal enzyme levels in cultured fibroblasts.
