for Hitachi High Speed Amino Acid Analyzer【W(wǎng)arning and Precautions】This kit is for research use only.Measurement results annot be used for diagnostic purpose.In amino acid analysis with an auto analyzer, the ninhydrin methodusing column chromatography and color reaction with ninhydrinfollowed by spectrophotometry is commonly adopted.This ninhydrin reagent kit is designed exclusively for Hitachi HighSpeed Amino Acid Analyzer.【Kit Contents】 (for 2 L)(Kit-1) Ninhydrin Solution ??????????????? 1 L x 1 bottle-ConteinsNinhydrinSodium borohydridePropylene glycol monomethyl ether(Kit-2) Buffer ??????????????????????? 1 L x 1 bottle-ContainsLithium Acetate DihydratePropylene glycol monomethyl ether【Principle of measurement】Inject a sample into a column filled with an alkaline formcation-exchange resin. Then, gradually increase the pH of eluentto dissolve out amino acids beginning with the highest acidity.Prepared Ninhydrin Reagent is then added to the eluate and heatedto generate colored compounds with wavelength of maximumabsorption at 570 nm for ɑ-amino acids and 440 nm for imino acids.Concentration of each amino acid is determined by comparing thepeak area of sample compound to that of the standard solution.【Operation instruction】(1) Preparation of Ninhydrin ReagentFor the model L-8900, L-8800, L-8500A and L-8500, nopreparation is necessary. Follow the instruction manual of theanalyzer to set Ninhydrin Solution (Kit-1) and Buffer (Kit-2).(2) Preparation of other reagentsMCI Buffer (Elution Buffer):Prepare, separay, and follow the manual of the analyzer touse.Amino Acid Standard SolutionPrepare separay, and follow the package insert of thestandard solution to use.(3) Protocol<Standard protocol>Inject the target sample to a column and elute with severaldifferent types of elution of citrate buffer solutions.Mix the Ninhydrin Reagent with the eluate and heated to emitcolor. At 570 nm and 440 nm, the absorbance values(integration value) are recorded automatically.Repeat the same procedure for the amino acid standard solutionto determine the absorbance value (integration value), andcompare each area to those taken using the sample (integrationvalue) to calculate a concentration of different amino acids. Thebase-line correction conducted at where no peak is shown.<Application of the model L-8900, L-8800, L-8500A, and L-8500Hitachi High Speed Amino Acid Analyzer>Inject 50 μL of test sample to the sulphonated polystyrene resinembedded column, run elution buffer at constant flow rate (0.4mL/min) by using a pump. Mix ninhydrin solution to the eluatefrom the column at constant rate (0.4 mL/min) and run it througha heated Teflon coil (In case of L-8900 and L-8800, use thereaction column, which is heated for 1.78 minutes) enclose it inflow cell of chromatometer. Measure absorbance value atwavelength of 570 nm and 440 nm, and record it with an autorecorder.Compare the peak area of an amino acid standard solutionachieved by the same procedure to the peak area of sample tocalculate the concentration in the test sample.Method of operation differs by the model.
